1) heroin doesn't calm people down.
2) it's much too slow acting to stop any sort of threat.
3) with any sort of tranquilizing drug there's a very fine line between an effective dose and a lethal dose, and that's assuming that you know the target's bodyweight. Factor in that some potential targets may have various degrees of tolerance to drugs and you're basically rolling the dice every time you shoot someone with a tranq. Might do nothing, might be lethal. The chance of the dose being in the goldilocks zone is virtually zero.
Do the people using tranqs in zoos and game reserves and stuff have the payload drug volume prescribed by the vets that work with those animals? Do they have different tranq loads for each animal?

Usually the person doing the tranqing IS a vet. They know the weight of the animal from the zoo's records so they have a good idea of how much drug to use.
I watched a ton of nature videos as a kid and sometimes they'd have to dart an animal multiple times several minutes apart if they didn't get the dose right the first time.

You can only hit one person with a single dart. A couple of m-80 firecrackers at the bottom of a coffee can filled with granulated fent can turn a riot into the spectator event of the year.
