Hitting them in the head gives them a head injury. Depending on all of the variables, they could have a headache with some cosmetic damage, a mild concussion, a severe concussion... strong potential for permanent brain damage.
The odds of a head strike immediately knocking someone out without permanent damage are extremely low.
If you're hitting them in the head enough to drop them it's extremely likely you've broken something or killed them.
There isn't a magic off switch, chemical or physical. Even stun guns have a significant fail rate.

I don't even know what they use for animal tranquilizers but there are plenty of drugs that would make someone dopey enough that you could very easily wrestle them into handcuffs. Wouldn't necessarily knock them out, but they wouldn't put up much of a fight either.
Benzodiazepines like Xanax can very easily do this. Their lethal dosage is also high enough that you're very unlikely to kill your victim by mistake.

When you fall unconscious all your muscles relax. Somebody who is sat back in a chair while unconscious can be seriously injured because their body weight compresses their blood vessels, effectively causing a crush injury
If somebody in a harness (eg working at heights) falls unconscious the absolute first thing you have to do before even trying to get them down is make them go horizontal, otherwise the blood vessels are compressed and the blood in their legs goes rancid.
Same reason why if you encounter a crush injury (eg person pinned against a wall by a car) you leave them there. They have to be removed professionally and get specialist treatment immediately, so that basically means the hospital has to come to them before they're freed.
Basically, don't knock someone out unless you're prepared to either cripple or kill them. The chances of them having no problems whatsoever are really low.

Youre talking about applying anesthesia on the fly with minimal knowledge on the patient. Pretty much youre liable to either dose low and end up in the embarrassing situation of the anesthetia just making them drowsy without knocking them out or knocking them out quickly and killing them. Knocking somebody out quickly without killing thems a lot easier said then done in a clinical setting let alone a resisting individual which is why anesthetists are some of the best paid medical professionals out there with the highest malpractice insurance. In any situation where you could get a highly trained doctor operator to simultaneously grapple and perfectly administer anesthesia youd be a fucking mouthbreathing retard not to just get two guys in there instead and hogtie the fucker with way more surety and lower chance of killing them on accident.
