Alright well whatever your weird weapon is, if it really goes through a solid one inch thick piece of wood it is probably going to penetrate human flesh and produce a serious wound. Like with any wound, it might not kill in one shot, so if you're defending yourself be aware of that.
if you're developing a weapon in secret you should remember there are plenty of simulated real human skulls out there, sleeping under bridges or having a cocaine seizure on the sidewalk, ripe for testing. The only difference is they're not attached to real people so its safe to test on.
Try it out on those skulls to see if your weapon would penetrate a real human skull later.

Your weapon will probably penetrate flesh but you have to remember that even normal bullets often don't kill on the first shot. Be prepared to shoot again fast. If your weapon is single shot maybe you should carry two.
Skull is definitely stronger then old rusty barrel. I suggest u buy pig or goat head and shoot it like the other anon suggests.
If it penetrates it will most likely penetrate your high-school bully too.

.22lr will penetrate 3-4 pine 2x4s, if you can go through that, I'd say it's lethal with good shot placement.
